Pengimplementasian Sistem Penjaminan Mutu Internal, knowledge merupakan sebuah aset penting yang dimiliki oleh instansi perguruan tinggi. Knowledge adalah keseluruhan bagian dari pengetahuan yang ada dan keterampilan individu yang digunakan untuk memecahkan masalah. Knowledge Management System dapat menjadi solusi untuk mensosialisasikan, mendokumentasikan, mendistribusikan serta mengatasi hilangnya knowledge serta dapat menjadi media untuk meningkatkan budaya sharing knowledge pada Universitas Mercu Buana. Metode perancangan aplikasi yang digunakan oleh penulis adalah metode Rational Unified Process (RUP) serta metode konversi knowledge menggunakan metode SECI model. Pada tahap akhir akan dilakukan pengujian aplikasi menggunakan metode blacbox user acceptance test, metode pengujian sistem menggunakan FGD serta menggunakan model pengukuran perangkat lunak adaptasi ISO 9126. Penelitian ini menghasilkan sebuah KMS yang dapat berfungsi untuk membantu mensosialisasikan knowledge, membangun budaya sharing knowledge, mendokumentasikan tacit maupun explicit knowledge serta dapat mendistribusikan dan mengatasi hilangnya knowledge dengan hasil pengujian blackbox testing user acceptance test meraih hasil yang baik yakni penerimaan 100% terhadap aplikasi KMS dan empat karakteristik model ISO meraih hasil 83.82% dengan kriteria baik.
